Why does LipoGalen® set new standards for excipients?

With LipoGalen®, Nordmann-Partner IOI Oleo introduces a new generation of functional, lipid-based excipients based on polyglycerol esters for the healthcare industry. The product family combines defined molecular structures with high stability and opens up new possibilities in pharmaceutical development.

Stability without polymorphism

LipoGalen® was developed from a deep understanding of the evolving challenges in pharmaceutical formulation. Unlike conventional triglycerides, the product crystallizes in a stable form and exhibits no polymorphism. Additionally specialized grades will show wateruptake and wettability enabling compatibilities with active ingredients that previously required use of surfactants and solubilizers to modify the inherent properties of lipid-based excipients.

What makes IOI Oleo’s LipoGalen® product family so interesting?

The following properties speak for themselves:

  • Sustainably sourced from natural raw materials
  • Plant-based
  • A family of multifunctional excipients
  • Custom-designed molecules
  • Free of mineral oil
  • Manufactured in cGMP-compliant facilities
  • A stable form without polymorphism
  • Defined molecular structures

Flexible Plattform („Chemical Toolbox“)

The product family offers a high degree of versatility, with over 200 possible molecular variants in theory. The result is targeted, application-specific solutions for modern formulations.

What makes it special is that key parameters can be adjusted:

  • HLB value (3–14)
  • Melting behavior (≤ 80 °C)
  • Wettability and water absorption

LipoGalen® 144

The product was launched as the first product in this new product line. Its physicochemical properties have been optimized for use as an innovative base for suppositories and ovules. LipoGalen® 144 incorporates hydrophilic and lipophilic active ingredients in established concentrations without the need for solubilizing or suspension-stabilizing additives. Another advantage is its high physical stability: Melting points remain stable over time, and even after months of fluctuating temperature cycles, the ovules and suppositories retain an attractive, glossy surface without any fat bloom forming. In a user panel study, lactic acid ovules based on LipoGalen® 144 demonstrated performance comparable to that of a leading medical device for women’s health. Additional applications in the field of rectal or vaginal health products, medical devices, and pharmaceutical dosage forms are currently in development.

Numerous Applications

LipoGalen® has a wide range of applications:

  • Controlled-release tablets: Matrix systems that enable precise control of the release profile throughout the entire shelf life—thanks to the superior properties of LipoGalen® compared to traditional triglycerides.
  • Solubilization and improvement of bioavailability: Ideal for hot-melt extrusion, improves the performance of poorly soluble active ingredients, and enables lower processing temperatures compared to established polymers in this application area.
  • Taste masking: Effective via hot-melt coating, improves patient acceptance.
  • As a lubricant and release agent in tableting: Optimized for continuous manufacturing processes.
  • Dermal formulations: Paraffin-free, supports clean-label and skin-friendly solutions.
  • Advanced drug delivery systems: Suitable for lipid nanoparticles and nanoemulsions, including inhalation therapies.
  • 3D printing: Compatible with melt extrusion techniques for personalized medicine and innovative dosage forms.

New Application Technologies

The following technologies are currently under investigation:

  • Compressed tablets
  • Multicomponent systems
  • Applications in the field of targeted drug delivery
  • Rapid-disintegrating tablets and films
  • Nano-lipid-colloid systems
  • Personalized dosage forms
  • Functional coatings for modified release
  • Semi-solid/liquid skin applications
  • Needle-free injection devices

Summary

LipoGalen® is a new class of lipid-based excipients with a defined molecular structure composed of high-purity polyglycerols and modern fatty acids. They exist as a stable, crystalline solid without polymorphism and are produced from sustainable raw materials. Their customizable properties include targeted hydrophilicity/lipophilicity (HLB), good wettability and water absorption, as well as use as a solubilizing agent with a low melting temperature — making them a multifunctional class of excipients.
